The Synthesis of 3-Nitrooxypropanol, a Methane Mitigant for Sustainable Farming with Environmental Impact: A Review

Abstract

Herein it is summarized and critically discussed the different methods reported in the current literature and patents for the synthesis of 3-nitrooxypropanol (3-NOP). Among the small molecules 3-NOP is possibly one of the smallest (chemical formula: C₃H₇NO₄; molecular weight: 121.09) showing interesting biological properties. Particularly, 3-NOP is an efficient inhibitor of methyl CoM reductase, an enzyme that catalyzes the last step of methanogenesis in anaerobic systems (e.g., ruminants), and a potent methane mitigant, able to reduce greenhouse gas emissions and climate change. 3-NOP is the active ingredient of Bovaer, a feed additive for ruminants. Furthermore, 3-NOP has been incorporated in a number of nitrate esters as NO-donors, showing diverse pharmacological activities for the treatment of inflammation or cardiovascular diseases.

• 5b To a solution of silver nitrate (5.34 g, 31.11 mmol) in acetonitrile (100 ml) was added via syringe 3-iodo-propan-1-ol (2.6 ml, 4.90 g, 26.33 mmol). The reaction mixture was stirred at room temperature for 48 h, then filtered over Celite. The filtrate was concentrated under reduced pressure and the residual oil was purified by flash chromatography (Petroleum Ether/EtOAc 7:3) yielding 3-hydropropyl nitrate as a brown oil (3.02 g, 24.95 mmol, 94%)…; R_f 0.20 (Pet. The oil is used as it is…3-Nitrooxypropan-1-ol can also be prepared from 1,3-propanediol (0.022 mmol) dissolved in ethyl acetate (50 ml), adding, at 0 °C, HNO₃ at 65% (w/v) (0.016 mmol, 1.55 ml), (CH₃CO)₂O (0.059 mmol, 5.56 ml), glacial AcOH (0.057 mmol, 5.56 ml). It is left to react at RT for 20 h. Then it is neutralized with 20% aqueous KOH and it is extracted with DCM, it is washed with H₂O, and dried, the solvent is concentrated and the residue is purified by silica chromatography EtOAc/hexane (1:3).

• 24b A solution of 3-bromo-1-propanol (5.42 g, 39.0 mmol) in acetonitrile (20 mL) was added to a solution of AgNO₃ (10.16 g, 59.8 mmol) in acetonitrile (50 mL) and stirred at room temperature for 24 h. To the reaction mixture was added brine (350 mL), and the mixture was stirred for 1 h. The silver salts were filtered off through Celite, and the filtrate was extracted with Et₂O (200 mL × 3). The organic layer was washed with brine, dried over Na₂SO₄, filtered, concentrated, and dried under vacuum to give the title compound (4.08 g, 86% yield, >95% purity) that was used in the next step without purification.

A procedure for 4-(nitrooxy)butan-1-ol was given therein as follows:
• 27b To a solution of 4-bromobutyl acetate (194 mg, 1 mmol) in acetonitrile (5 mL) was added a solution of AgNO₃ (510 mg, 3 mmol) in acetonitrile (5 mL), and the mixture was then kept stirring at reflux for 24 h. Then, the reaction mixture was added to brine (25 mL) and was stirred for another 1 h. The silver salts were filtered off through Celite, and the filtrate was extracted with Et₂O (40 mL × 3). The organic layer was washed with brine, dried over Na₂SO₄, filtered, concentrated, and dried under vacuum. The crude 4-(nitrooxy)butyl acetate was dissolved in a solution of 2 N NaOH (0.5 mL, 1 mmol) in MeOH (5 mL) and was stirred at room temperature for 2 h. The reaction mixture was acidified with 3 N HCl (aq.) and then concentrated under reduced pressure. The residue was partitioned between 3 N HCl (aq.) and CH₂Cl₂. The organic extract was washed with water and brine, dried over Na₂SO₄, filtered, and concentrated under vacuum. Then, the crude product was purified by flash chromatography on silica gel (eluted with 0–40% ethyl acetate in hexane) to provide the title compound as a yellowish oil (95 mg, 70% yield).

• 34b Concentrated nitric acid [22 mmol], glacial acetic acid [5 mL], and acetic anhydride [5 mL] were consecutively added to an ice-cold solution of the appropriate diol [20 mmol] in 50 mL ethyl acetate under nitrogen... The reaction mixture was stirred at room temperature overnight, and the solution was washed with a 10% aqueous solution of sodium bicarbonate. The organic layer was dried over potassium carbonate, filtered, and concentrated under reduced pressure. The oily residue was purified by flash chromatography, eluting with petroleum ether-ethyl acetate.

In this article, Rekka and co-workers also reported the synthesis of 3-NOP from 3-chloropropan-1-ol in 40% yield as follows:
• 34c Silver nitrate [35 mmol] was added to a solution of the corresponding 2-chloroethanol or 3-chloropropanol [23 mmol] in acetonitrile [40 mL]. The reaction mixture was stirred at room temperature overnight and was light protected. The reaction mixture was filtered and the volatiles were removed under reduced pressure. The residue was dissolved in diethyl ether and washed with saturated NaCl solution. The organic layer was dried over calcium chloride, filtered, and concentrated. The obtained oil was purified by flash chromatography, eluting with petroleum ether-ethyl acetate.
