Abstract

An ultramicro method for the investigation of the transport and turnover of tissue nucleotides in vitro is described. The method was used to demonstrate that the incorporation of adenine-8-3H into the nucleotides of human leukocytes was linear during one hour of incubation. The incorporation was strongly inhibited by caffeine. The usefulness of the method was demonstrated also by showing that pregnant mare serum gonadotropin (PMSG) caused a five-to ten-fold increase in the cyclic AMP content of hamster ovaries either in the presence or in the absence of caffeine. PMSG also increased the amount of label in the ATP pool; this effect was inhibited by caffeine.
