Effect of a Gold-Chloroquine Complex on Inflammation-related Leukocyte Functions and Cell Viability

Summary
This study evaluates a new gold-chloro-quine complex [hexafluorophosphate triphenylphosphine chloroquine gold (I), Au(CQ)(PPh3)PF6, referred to hereinafter as CQAu] in terms of its anti-inflammatory and toxic effects on immune cells compared to auranofin (AF). CQAu and AF were compared for their effects on a) tumor necrosis factor-α (TNF-α) and in-terleukin-6 (IL-6) release by human lymphocytes and murine macrophages, b) functionality and survival of polymor-phonuclear leukocytes (PMN), c) PMN function in the presence of human serum albumin as a competitive inhibitor, d) mitogen-induced proliferation of human lymphocytes and e) TNF-α and IL-6 response of mice to lipopolysaccharide (LPS). CQAu was found to be generally similar to AF, or somewhat less effective, in terms of its activity in different assays of human immune cell function. For several of the assays, this was related to the greater cytotoxicity of AF. However, the two drugs did show comparable inhibitory effects on TNF-α and IL-6 production in a mouse model in vitro and in vivo, which were independent of a cyto-toxic effect.
CQAu merits further investigation as a gold drug with potential applications in treating inflammatory disease.
Zusammenfassung
Wirkung eines Gold-Chloroquin-Komple-xes auf entzündungsbezogene Leukozyten-Funktionen und Überlebensfähigkeit von Zellen / Vergleich mit Auranofin
Die vorliegende Studie evaluiert einen neuen Gold-Chloroquin-Komplex [Hexa-fluorphosphat-triphenylphosphin-chloro-quin-Gold (I), Au(CQ)(PPh3)PF6; nachfolgend CQAu genannt] im Hinblick auf entzündungshemmende und toxische Wirkungen auf Immunzellen im Vergleich zu Auranofin (AF). Es wurde die Wirkung von CQAu und AF a) auf den Tumorne-krosefaktor alpha (TNF-α) und die Freisetzung von Interleukin-6 (IL-6) aus menschlichen Lymphozyten und Maus-Makrophagen geprüft, b) auf die Funktionalität und das Überleben von polymorph-kernigen Leukozyten (PMN), c) auf die die Funktion der PMN in Gegenwart von menschlichem Serumalbumin als kompetitiver Inhibitor, d) auf die Mitogen-aktivierte Vermehrung menschlicher Lymphozyten sowie e) auf die Reaktion von TNF-α und IL-6 (in Mäusen) auf Lipopolysaccharid. CQAu war im allgemeinen ähnlich wirksam oder etwas weniger wirksam hinsichtlich seiner Aktivität in verschiedenen Tests zur Funktion humaner Immunzellen. In mehreren Tests war dies auf die gröβere Zytotoxizität von AF zurückzuführen. Die beiden Substanzen zeigten vergleichbare Hemmwirkung auf die Produktion von TNF-α und IL-6 in einem Mausmodell in vitro und in vivo, und zwar unabhängig vom zytotoxischen Effekt. CQAu sollte im Hinblick auf seine potentielle Verwendbarkeit bei entzündlichen Krankheiten weiter untersucht werden.
