In vitro Antifungal Properties, Structure-activity Relationships and Studies on the Mode of Action of N-Phenyl, N-Aryl, N-Phenylalkyl Maleimides and Related Compounds

Summary
The synthesis, in vitro antifungal evaluation and structure-activity relationship studies of 14 compounds of the N-phenyl-, N-aryl-, N-phenylalkylmaleim-ide and 3,4-dichloromaleimide series are reported. The compounds were evaluated against a panel of standardized yeasts and filamentous fungi as well as clinical isolates of Candida albicans. The activities of N-phenylalkyl-3,4-dichloromaleimide derivatives but not those of N-phenylalkylmaleimide derivatives showed to be dependent on the length of the alkyl chain. N-Phenylpropyl-3,4-dichloromaleimide showed the broadest spectrum of action and lower minimal inhibitory concentrations (MIC) in all of the fungi tested. The nitrogen-carbon distance between the two rings seems to play an important role in the antifungal behavior of these compounds. The most active structure showed inhibited (1,3)β-D-glucan and chitin synthases, enzymes that catalyze the synthesis of the major fungal cell-wall polymers.

Antimykotische Eigenschaften in vitro, Struktur-Wirkungs-Beziehungen und Studien zum Wirkungsmodus von N-Phenyl-, N-Aryl- und N-Phenylalkyl-Maleimi-den sowie verwandter Verbindungen
Es wird über die Synthese, antimykotische Wirkung in vitro sowie Struktur-Wirkungs-Beziehungen von 14 Verbindungen der N-Phenyl-, N-Aryl-, N-Phenylalkyl-Maleimid- und 3,4-Dichloro-Maleimid-Serie berichtet. Die Verbindungen wurden an standardisierten Hefen und Fadenpilzen sowie klinischen Isolaten von Candida albicans auf Wirksamkeit geprüft. Die Wirkung von N-Phenylalkyl-3,4-dichloro-maleimiden hing von der von der Länge der Alkyl-Kette ab, was bei den N-Phenylalkyl-maleimid-Derivaten nicht festzustellen war. Die N-Phenylpropyl-3,4-dichloro-maleimide zeigten bei allen getesteten Pilzen das breiteste Wirkungsspektrum und die niedrigste minimale Hemmkonzentration (MHK). Der
Abstand zwischen dem Stickstoff- und Kohlenstoffatom zwischen den beiden Ringen scheint eine wichtige Rolle im antimykotischen Verhalten dieser Verbindungen zu spielen. Die wirksamste Struktur hemmte (1,3)β-D-Glucan- und Chitin-Synthasen, Enzyme, welche die Synthese des wichtigsten Polymers der Pilzzellwand katalysieren.
