In vitro Activities of Thiadiazine Derivatives against Leishmania amazonensis

Summary
Ten thiadiazine derivatives were tested in vitro for antiparasitic effects against both extracellular promastigotes and intracellular amastigotes of Leishmania amazonensis. The results showed that the evaluated compounds exhibited a strong antiproliferative activity on all developmental stages of the parasite. The minimal inhibitory concentration and the 50 % effective concentration values against the promastigote were 2.1-5.1 µg/ml and 0.6-1.8 µg/ml, respectively. The tested compounds caused an irreversible inhibition of the promastigote growth either after 1 h of treatment with 10 µg/ml or after 24 h with 1 µg/ml. Also, the thiadiazine derivatives were active against amastigotes producing between 12 and 89 % of reduction of infection at 100 ng/ml. However, the compounds exhibited high toxicity and provoked inhibition of the phagocytosis in the murine host cell.
Zusammenfassung
In-vitro-Wirkung von Thiadiazin-Derivaten gegen Leishmania amazonensis Zehn Thiadiazin-Derivate wurden in vitro auf antiparasitäre Wirkung gegen Leishmania amazonensis-Promastigoten (extrazellulär) und -Amastigoten (intrazellulär) untersucht. Die Ergebnisse zeigten, daβ die untersuchten Verbindungen die Vermehrung der Parasiten in allen Entwicklungsstadien stark hemmten. Die minimale Hemmkonzentration und die EC50-Werte betrugen bei den Promastigoten 2,1-5,1 µg/mL bzw. 0,61-1,8 µg/mL. Eine Hemmung des Wachstums der Promastigoten wurde nach einstündiger Behandlung mit 10 µg/mL bzw. nach 24stündiger Behandlung mit 10 µg/mL beobachtet. Die Thiadiazin-derivate waren auch gegenüber den Amastigoten wirksam, wobei sie bei einer Konzentration von 100 ng/mL die Infektion um 12 bis 89 % reduzierten. Allerdings erwiesen sich die Verbindungen als hochtoxisch und hemmten die Phagozytose in Wirtszellen der Maus.
