Chemical profile of Justicia gendarussa a medicinal plant used for male contraception

Justicia gendarussa Burm. F. (Acanthaceae) is a medicinal plant used to reduce male fertility among the Papuan tribes in Indonesia [1]. Traditional water decoction and the pills containing standardised extract used in clinical trials were analysed by HPLC-PDA-ESI-MS and UHPLC-TOF-HRMS for chemical profiling. Using this approach several 2-aminobenzyl derivatives and two flavonoids were dereplicated. UV and HRMS data also suggested the presence of an additional new 2-aminobenzyl derivative. In order to quantify these compounds in the various extracts and identify the new constituent, an efficient single step medium pressure liquid chromatography (MPLC-UV) was performed. The seven main constituents were isolated and as expected one a new 2-aminobenzyl derivative was identified it structure being confirmed by NMR, and HRMS. Isolated standards were used to develop an HPLC-UV quantification method. The 2-aminobenzyl derivative and flavonoid content of the traditional water decoction was compared with the standardised extract used in clinical trials. This revealed that pills used in clinical trials contain mainly the flavonoid gendarusin A, hypothesised active compound, and that the 2-aminobenzyl derivative were significantly removed by the standardisation process. Comparison of various J. gendarussa specimens collected in different regions of Indonesia was useful for the selection of plant material rich in gendarusin A.
