According to absorption spectra, at least four different 99Tc-HEDP complexes [complex I (colorless), complex II (yellow), complex III (pink-red) and complex IV (brown)] were observed under various labeling conditions. Both pH and Sn (II)-HEDP concentration greatly influenced the formation of these complexes. In acidic or neutral media, complex II was the main product while in alkaline media complex III and complex IV were formed below concentration levels of 1 x 10–3M of Sn (II) and of 3x 10–3M of HEDP. At high Sn (II)-HEDP concentrations complexes I and II were formed, while complexes III and IV were found at lower Sn (II)-HEDP concentrations in weak alkaline media. All four 99(m)Tc-HEDP complexes were stable against air oxidation until at least 6 hrs after preparation. Complexes I and II were stable against dilution by distilled water, while complexes III and IV were unstable and readily dissociated. In organ distribution studies high soft-tissue uptakes were observed with complexes III and IV, while complexes I and II had high bone-seeking properties.
Nach Aussage der Absorptionsspektren wurden bei unterschiedlichen Markierungsbedingungen zumindest 4 verschiedene 99Tc-HEDP-Komplexe gebildet und zwar: Komplex I (farblos), Komplex II (gelb), Komplex III (rosarot) und Komplex IV (braun). Die Bildung dieser Komplexe hängt stark sowohl vom pH-Wert als auch von der Sn (II)-HEDP-Konzentration ab. In saurer und neutraler Lösung ist der Komplex II das Hauptprodukt; in alkalischer Lösung bei Sn-II-Konzentrationen unter 1×10– 3M und bei HEDP-Konzentrationen unter 3 ×10– 3M werden die Komplexe III und IV gebildet. Bei hohen Sn (II)-HEDP-Konzentrationen entstehen die Komplexe I und II. Die Komplexe III und IV wurden bei niedrigen Sn (II)-HEDP-Konzentrationen im schwach alkalischen Medium gefunden. Alle vier 99(m)Tc-HEDP-Komplexe waren für mindestens 6 Std. nach der Herstellung stabil gegen Oxydation durch Luft. Die Komplexe I und II zeigten sich unempfindlich gegenüber einer Verdünnung mit destilliertem Wasser, die Komplexe III und IV waren jedoch nicht stabil gegenüber Verdünnung mit Wasser und dissoziierten leicht. Beim Studium der Organverteilung zeigte sich, daß weiche Gewebe große Mengen der Komplexe III und IV aufnahmen, während sich in den Knochen die Komplexe I und II stark anreicherten.
