Optimal conditions of HSA aggregation in acetate buffer (pH = 4.6) were investigated. With the aim of obtaining a higher percentage of fixation in the lungs, the aggregates were rinsed three times with acetate buffer. A MAA suspension was lyophilized with SnCl22H2O (10 μg/ml) solution. The lyophilized MAA was then labelled by adding pertechnetate of the desired activity in a volume of up to 5 ml. The radiochemical purity of 99mTc-MAA, greater than 95%, was determined by the methods of thin layer chromatography and low-voltage electrophoresis. All MAAs were uniform in shape whereas the size varied between 20 and 100 μm.

Biodistribution studies have shown the maximum radioactivity to be deposited in the animals' lungs immediately after application of 99mTc-MAA.

Es wurden die zur Aggregation von HSA in Azetatpuffer (pH 4.6) nötigen Optimalbedingungen untersucht. Zwecks Erreichung eines höheren Fixierungsgrades in der Lunge wurden die Aggregate dreimal mit Azetatpuffer gewaschen. Die MAA-Suspension wurde mit einer Lösung von SnCl22H2O (10 μg/ml) lyophilisiert.

Das Markierungsverfahren für lyophilisierte MAAs besteht in der Zugabe von Pertechnetat in der gewünschten Aktivität in einem Volumen bis zu 5 ml.

Die radiochemische Reinheit vom 99mTc-MAA ist höher als 95 % und wurde mittels Dünnschicht-Chromatographie und Niederspannungs-Elektrophorese bestimmt. Die MAAs waren uniform in ihrer Form während die Größe zwischen 20–100 μm schwankte.

Durch Untersuchungen der Bioverteilung wurde festgestellt, daß die maximale Aktivität in den Lungen der Versuchstiere unmittelbar nach der Applikation von 99mTc-MAA deponiert wird.
