Studies in Nitrogen-Cold Stabilized Polyphosphates

Wir präsentieren ein neues Verfahren für die Vorbereitung von PPO₄-Vorrichtungen, die sich für ^99mTc-Kennzeichnung eignen. Die von uns vorgeschlagene Methode benutzt die N₂-Stabilisierung der Polyphosphat-Vorrichtung anstelle der Trockenfriermethode, über die berichtet wurde. Die auf diese Art vorbereiteten (N₂-stabilisierten) Polyphosphat-Vorrichtungen zeigten eine chromatographische Bindungseffizienz zwischen 87 und 96% bei der Benutzung von Dünnschicht-Chromatographie mit verschiedenen Lösesystemen. In-vivo-Studien an Tieren zeigten für diese Methode eine 6wöchige Stabilität. Auch erscheint eine sehr gute Korrelation zwischen in-vitro-Bindeeffizienz und in-vivo-Wirksamkeit zu bestehen. Nachweislich kann PPO4 auf intravenösem, intramuskulärem oder intraperitonealem Wege zugeführt werden, wobei der Qualitätsunterschied der Studien gering ist.

Summary

A new procedure for the preparation of PPO kits suitable for ^99mTc labeling has been presented. Our proposed method utilizes N2 stabilization of the polyphosphate kit rather than the reported freeze dry method. The polyphosphate kits prepared in this manner (N2 stabilized) have demonstrated a chromatographic binding efficiency ranging from 87 to 96% using TLC with different solvent systems. In vivo studies in animals have exhibited a six week stability for this method. Also, there appears to be a very good correlation between invitro binding efficiency and in vivo efficacy. There is evidence that PPO may be administered by intravenous, intramuscular or intraperitoneal routes with little difference in quality of the studies.

Un procédé nouveau permettant d’obtenir des préparations de PPO₄ convenant au marquage par le ^99mTc a été présenté. La méthode que nous proposons utilise la stabilisation au N₂ de la préparation de polyphosphates, plutôt que la méthode de dessication au froid décrite. Les préparations de polyphosphates ainsi obtenues (stabilisées au N₂ ) ont révélé une capacité de fixation chromatographique variant de 87 à 96% en utilisant le méthode de Chromatographie à couches minces (TLC) avec divers systèmes dissolvant. Les études in vivo chez les animaux ont démontré, par cette méthode, une stabilité de six semaines. De plus, il semble y avoir un très bon rapport entre la capacité de fixation in-vitro et l’efficacité in vivo. Il est prouvé que PPO₄ peut être administré par voie intraveineuse, intramusculaire ou intrapéritonéale sans que la qualité des études varie considérablement.
