9-(2′-Deoxy-2′-C-ethynyl-β-d-arabino-pentofuranosyl)-guanine phosphoramidite (8) has been prepared from guanosine in 9 steps and incorporated into oligodeoxynucleotides. Substitution of two or three ethynyl-modified guanosines for deoxyguanosine within d(CG)3 or d(GC)3 leads to a Z-DNA-like conformation of the resulting duplex regardless of salt concentration.
