Effects of an Active Substance Isolated from the Roots of Angelica shkiokiana on Leukotriene and Monohydroxyeicosatetreaenoic Acid Biosyntheses in Human Polymorphonuclear Leukocytes

Abstract
It was found that the EtOAc fraction of the roots of Angelica shkiokiana Makino inhibited calcium ionophore-induced LTB4 and LTC4 formations in human polymorphonuclear leukocytes (PMN-L). And the active substance isolated from this root of was elucidated to be 3′ (R), 4′ (R)-3′-epoxyangeloyloxy-4′-acetoxy-3′, 4′-dihydroseselin (YN-1). Furthermore, YN-I at concentrations of 4-400 µg/ml inhibited calcium ionophore-induced 12-HETE, 5-HETE, LTB4, and LTC4 formation in human PMN-L, dose-dependently, as shown by measurements with reversed phase HPLC and radioimmunoassay systems.
