Summary
Creutzfeldt-Jakob disease (CJD) is a neurodegenerative disorder caused by a transmissible prion protein (PrP). Prions may be both inherited and infective and are believed to be composed largely, if not entirely of an abnormal isoform of cellular prion protein (PrPc) designated prion protein scrapie (PrPSc). The PrP gene controls synthesis, replication, incubation of prions and the topology of tissue alterations, although the precise nature of infection and protein transformation remains unknown. Several PrP gene mutations feature in hereditary human prion diseases. Iatrogenic CJD is a serious complication, but the occurrence of CJD following exposure to prions associated with scrapie or BSE seems very unlikely.

Die Creutzfeldt-Jakob-Krankheit (CJK) ist eine durch Prion-Protein hervorgerufene neurodegenerative Erkrankung. Die Erkrankung kann sowohl genetisch als auch infektiös verursacht sein. Das Prion besteht zum größten Teil, wahrscheinlich gänzlich aus einer abnormen Scrapie Prion-Proteinisoform (PrPSc) des zellulären Prion-Proteins (PrPc). Synthese, Replikation, Inkubation des Prions sowie die Topologie der Gewebeveränderungen werden durch das PrP-Gen kontrolliert. Die genaue Natur der Infektion und der Proteintransformation ist allerdings weiterhin unklar. Verschiedene PrP Genmutationen charakterisieren humane Prionerkrankungen. Die iatrogene CJK stellt eine gravierende Komplikation dar; eine CJK nach Scrapie oder BSE Prion Exposition erscheint allerdings eher unwahrscheinlich.
