Abstract
The effects of didrovaltratum and of four degradation products of valepotriates were studied on the motor activity of light-dark synchronized mice. All compounds reduced spontaneous motility in a dose-dependent way when applied intraperitoneally during the dark phase. The respective ED50-values for valtroxal, 8,9-didehydro-7-hydroxy-dolichodial, 11-ethoxyviburtinal, baldrinal and didrovaltratum were 59, 78, 161, 300 and 131 µmol/kg body weight. The therapeutic index was greater for the degradation products than for didrovaltratum. After oral application either only a slight depressant (valtroxal, 11-ethoxyviburtinal) or no obvious dose-dependent effects were observed even after very high drug doses in the range of 120-760 µmol/kg body weight.
