Zusammenfassung
Interleukin-2 (IL-2) hat bei der Tumorabwehr durch eine Vielzahl an verstärkenden Wirkungen auf das Immunsystem eine wesentliche Bedeutung. Die Konzentration von IL-2 kann durch die Vitalität der Interleukin-2 abhängigen CTLL-Zellinie im Mikro-Elisa-System gemessen werden. IL-2 wurde aus mononukleä-ren Zellen dosisabhängig nach Stimulation mit Mitogenen wie Phytohämagglutinin A (PHA), Phorbolester (PMA) oder dem monoklonalen T-Zellantikörper OKT3 freigesetzt. Eine maximale Interleukin-2 Sekretion wurde nach gleichzeitiger Stimulation mit Phytohämagglutinin A und Phorbolester (PHA und PMA) erreicht. Als Kontrollgruppen dienten gesunde Probanden mit niedrigem oder hohem Alkohol- und Zigarettenkonsum. Bei Patienten mit einem Oropharynxkarzinom konnte eine signifikant niedrigere Interleukin-2 Sekretion im Vergleich zu Kehlkopfkarzinompatienten und zu gesunden Spendern beobachtet werden. Diese Beobachtung läßt eine direkte immunologische Beeinflussung durch das Plattenepithelkarzinom vermuten, welches im Oropharynx in unmittelbarer Nähe zum lymphoepithelialen Gewebe des Waldeyer'schen Rachenrings wächst.
Summary
The various strengthening influences of interleukin-2 (IL-2) on the immunological system is of intrinsic importance to tumour defense. IL-2 concentration can be measured by the determination of the IL-2 dependent CTLL cell line in a micro-Elisa-assay. The IL-2 secretion is dose dependent after stimulation with the mitogens phytohaemagglutinin A (PHA), phorbolester (PMA) or the monoclonal T cell antibody OKT3. Maximal IL-2 release is achieved by concomitant activation with phytohaemagglutinin A (PHA) and phorbolester (PMA). As controls healthy donors with low and high alcohol and cigarette consumption were used. In patients with cancer of the oropharynx, a significant reduction in the IL-2 secretion was observed when compared with the larynx cancer group or with healthy donors. This might suppose an immunological influence of the squamous cell carcinoma which grows in the oropharynx in close proximity to lymphatic cells and vessels which are numerously present in the tonsillar ring of Waldeyer.
