Abstract
Interleukin-1, tumor necrosis factor and interleukin-6 are mediators of inflammation, produced by monocytes/macrophages and a multitude of other cells. There is a causal connection with the pathogenesis of juvenile rheumatoid arthritis. All cytokines are characterized by pleiotrope paracrine, autocrine and endocrine effects on target cells.
This paper informs clinicians on the various and partly overlapping effects of the three cytokines mentioned above in relation to the persisting and self-perpetuating reactions of inflammation in juvenile rheumatoid arthritis. Additional mechanisms of regulation of the network of cytokines are described.
Zusammenfassung
Interleukin-1, Tumor-Nekrose-Faktor und Interleukin-6 sind an der Pathogenese der juvenilen chronischen Arthritis (jcA) ursächlich beteiligte Entzündungsmediatoren, die von Monozyten/Makrophagen und einer Vielzahl anderer Zellen synthetisiert werden. Sie üben pleiotrope parakrine, autokrine und endokrine Wirkungen auf ihre Zielzellen aus.
Diese Arbeit gibt dem Kliniker einen Überblick über die vielfältigen und sich zum Teil überlappenden Effekte der genannten Zytokine vor dem Hintergrund der jcA als dem Beispiel einer persistierenden und sich verselbständigenden Entzündungsreaktion und geht auf Regulationsmechanismen innerhalb des Zytokinnetzwerkes ein.
