Sequentielle Verabreichung von Methotrexat und Fluorouracil bei metastasierenden kolorektalen Karzinomen: Ergebnisse einer Phase-II-Pilotstudie

20 Patienten mit metastasierendem kolorektalem Karzinom wurden mit einer sequentiellen Kombination von Methotrexat und 5-Fluorouracil behandelt. Fünf Patienten waren bereits mit 5-Fluorouracil vorbehandelt. Während bei den chemotherapeutisch vorbehandelten Patienten zwei Teilremissionen, zwei Minimalremissionen und eine Progression auftraten, kam es bei den übrigen Patienten zu einer Vollremission, zu sechs Teilremissionen, fünf Minimalremissionen und zu drei Progressionen. Die mittlere Teilremissionsdauer bei diesen Patienten betrug 7,9 Monate, und die mittlere Überlebenszeit wird voraussichtlich über ein Jahr betragen. Die Toxizität der Behandlung war gering.

Abstract

A sequential combination of methotrexate and 5-fluorouracil was used for treatment of 20 patients with metastasizing colorectal carcinomas. Pretreatment with 5-fluorouracil had been given to five patients. While the chemotherapeutically pretreated patients showed two partial remissions, two minimal remissions and one progression, the remaining patients showed one full remission, six partial remissions, five minimal remissions and three progressions. The average partial remission time of these patients was 7,9 months. The average survival time will probably be more than one year. The toxicity of treatment was slight.
