Chemotherapieprotokoll zur Behandlung des metastasierenden Magenkarzinoms: Methotrexat, Adriamycin und 5-Fluorouracil

Zusammenfassung

In einer Phase-I/II-Studie wurde geprüft, inwieweit eine sequentielle zytostatische Kombination von Methotrexat, 5-Fluorouracil und Adriamycin bei Patienten mit metastasiertem Magenkarzinom wirksam ist. 30 Patienten mit einem Karnofsky-Index zwischen 40 und 70 % wurden bislang behandelt. Die Ansprechrate (komplette und partielle Remission) betrug ∼ 63 % (19 von 30 Patienten). Zwei von 30 Patienten kamen in eine komplette Remission. Die mediane Überlebenszeit der Patienten mit einer Remission ist zur Zeit noch nicht zu beurteilen: Etwa 68 % leben noch nach mehr als 17 Monaten. Die mediane Überlebenszeit der Patienten mit progredientem Tumor betrug nur 5 Monate. Der Unterschied der Überlebenskurven ist statistisch signifikant (P < 0,05). Die Studie wird fortgesetzt.

Abstract

The possible effectiveness of methotrexate, 5-fluorouracil and adriamycin was tested in a phase I/II study of patients with metastasising gastric carcinoma. Complete or partial remission occurred in 19 of 30 patients (∼ 63 %). Two of 30 patients had complete remission. Median survival time of patients with remission cannot as yet be determined: about 68 % are still alive after 17 months. Median survival time of patients with advanced tumour was only five months. The difference in survival curve is statistically significant (P < 0.05). The study is continuing.
