Structural Identification of Products from the Chloromethylation of Salicylaldehyde

In the functionalization of salicylaldehyde to give 5-(chloromethyl)salicylaldehyde, two byproducts [5-(hydroxymethyl)salicylaldehyde and 5,5′-methylenebis(salicylaldehyde)] were also isolated. Detailed characterizations and structural analyses of all three products by single-crystal X-ray diffraction, multinuclear NMR spectroscopy, high-resolution mass spectrometry, and IR spectroscopic techniques are presented and discussed. A strategy is presented for the preferential isolation of the two byproducts through column chromatography.

19 5-(Chloromethyl)salicylaldehyde [2; 5-(Chloromethyl)-2-hydroxybenzaldehyde] Salicylaldehyde (15 g, 0.1228 mol) and 30% aq formaldehyde (5 mL) were added to a stirred solution of concd HCl (100 mL) at 0 °C in an ice bath, and the mixture was allowed to warm to r.t. After 24 h, the mixture was again cooled to 0 °C, and the solid that precipitated out was washed with 5% aq NaHCO₃ (30 mL), collected by filtration, and dried in air. The resulting mixture (12 g) was dissolved in Et₂O (50 mL), and 2 was recovered by evaporation of the solvent from an open beaker in a fume hood. Further purification was achieved by washing with hexane and crystallization from Et₂O to give a white powder; yield: 10.8 g (0.0619 mol, 50%); mp 84–86 °C (lit.¹⁸ 85–86 °C). IR (ATR, air): 1649, 1581, 1476, 1283 cm^–1. ¹H NMR (400 MHz, DMSO-d ₆): δ = 4.75 (s, 2 H), 7.01 (d, J = 8.66 Hz, 1 H), 7.58 (dd, J = 8.51/2.30 Hz, 1 H), 7.72 (d, J = 2.29 Hz, 1 H), 10.26 (s, 1 H), 10.90 (s, 1 H). ¹³C NMR (100 MHz, DMSO-d ₆): δ = 45.71, 117.75, 122.17, 128.80, 129.12, 136.95, 160.73, 190.72. Anal. Calcd for C₈H₇ClO₂·⅛H₂O: C, 55.59; H, 4.23; Found: C, 55.29; H, 3.9.
20 5-(Hydroxymethyl)salicylaldehyde [3; 2-Hydroxy-5-(hydroxymethyl)benzaldehyde] and 5,5′-Methylenebis(salicylaldehyde) [4; 3,3′-Methylenebis(6-hydroxybenzaldehyde)] The same procedure as used in the preparation of 2 was followed. For the isolation of 3 and 4 in pure form, 12 g of the mixture was added to the top of a column packed with silica gel. Elution was started with pure hexane, and the solvent polarity was slowly increased to 30% EtOAc–hexane. Compound 4 eluted out first (R_f = 0.86), followed by 3 (R_f = 0.46) (EtOAc–hexane, 1:1). Each product was recovered by solvent evaporation from open beakers placed in a fume hood. 3 White powder; yield: 1.64 g (9%); mp 102–103 °C. IR (ATR, air): 3315, 3218, 2876, 1650, 1582, 1482, 1275, 1019 cm^–1. ¹H NMR (400 MHz, DMSO-d₆): δ = 4.42 [d, J = 4.02 Hz (s after D exchange), 2 H], 5.13 (s, exchangeable, 1 H), 6.95 (d, J = 8.58 Hz, 1 H), 7.46 (dd, J = 8.49, 2.25 Hz, 1 H), 7.60 (d, J = 2.10 Hz, 1 H), 10.24 (s, 1 H), 10.59 (s, 1 H). ¹³C NMR (100 MHz, DMSO-d₆): δ = 62.08, 116.99, 121.75, 127.09, 133.60, 135.00, 159.63, 191.84. HRMS (ESI^–): m/z [M – H] calcd: 151.0395; found: 151.0375. Anal. Calcd for C₈H₈O₃: C, 63.15; H, 5.30. Found: C, 63.46; H, 6.28. Single crystals suitable for studies by X-ray diffraction were grown from EtOAc layered with hexane. 4 White powder; yield: 3.31 g (11%); mp 135–138 °C. IR (ATR, air): 3057, 2859, 1648, 1588, 1480, 1273 cm^–1. ¹H NMR (600 MHz, DMSO-d₆): δ = 3.84 (s, 2 H), 6.93 (d, J = 8.45 Hz, 2 H), 7.37 (dd, J = 8.28/2.24 Hz, 2 H), 7.47 (d, J = 2.28 Hz, 2 H), 10.21 (s, 2 H), 10.58 (s, 2 H). ¹³C NMR (150 MHz, DMSO-d₆): δ = 38.62, 117.49, 122.10, 128.45, 132.29, 136.89, 159.24, 191.50.
