Phylogenie und Evolution von Hormonsystemen

Zusammenfassung

Nach der klassischen Definition sind Hormone Substanzen, die von endokrinen Drüsen- oder Nervenzellen sekretiert werden und über den Blutstrom oder andere Körperflüssigkeiten in einen anderen Teil des Körpers gelangen, wo sie in geringsten Konzentrationen physiologische Prozesse auslösen. Betrachtungen zum Urspung und zur Evolution von Zell-Zell-Kommunikationssytemen lassen vermuten, dass exokrine Pheromone (Stoffwechsel-verwertbare Moleküle, Toxine) als die ursprünglichen bioregulatorischen Signalmoleküle bei der chemischen Kommunikation von einzelligen Organismen untereinander und mit der Biosphäre angesehen werden können. Die weite Verbreitung und die strukturelle Diversität von Pheromonen macht es wahrscheinlich, das solche Moleküle und ihre Rezeptoren als Vorläufermodule der Zell-Zell-Kommunikation in Metazoen (Vielzeller) dienten. Neurosekretorische Zellen, wie sie bei Cnidariern vorliegen, waren wahrscheinlich die Grundmodule zur Entwicklung von neurohormonellen Systemen höherer Tiere. Untersuchungen der letzten Jahre an genetischen Modellorganismen, wie Drosophila oder der Maus, zeigen, dass chemische Kommunikation zwischen benachbarten oder im Körper weit entfernten Zellen nicht nur von spezialisierten endokrinen oder neurosekretorischen Zellen ausgeht, sondern dass auch Gewebe und Organe wie das Herz oder das Fettgewebe eine unerwartete endokrine Funktion aufweisen (z. B. der Leptin-Signalweg). Durch vergleichende funktionelle Studien konnte nachgewiesen werden, dass molekulare Komponenten der Hormonsysteme zelluläre Signalsysteme repräsentieren, die allgemein bei Zell-Zell-Kommunikationsprozessen und der Differenzierung von Zelltypen während der Ontogenese eine zentrale Rolle spielen. Manche ihrer Funktionen sind evolutionär konserviert, andere jedoch nicht, wie am Beispiel von Steroidhormonen und des Prolaktin-Signalweges deutlich gemacht wird.

Abstract

Classically hormones are defined as molecules that are secreted by endocrine glandular or neurosecretory cells into the blood stream and transported to their target tissue where they induce physiological processes at very low concentrations. Studies on the potential origin and the evolution of cell-to-cell communication systems suggest that exocrine pheromones (food signals and toxins) might have been the primitive bioregulatory molecules of unicellular organisms for chemical communication with each other and with the biosphere. The broad distribution and the structural diversity of pheromones suggests that these molecules and their receptors were predecessor modules of cell communication systems in metazoa. Neurosecretory cells, as we find them in Cnidarians, possibly served as basic modules for the evolution of neurohormonal systems of higher animals. Studies on genetic model organisms, such as Drosophila or the mouse, have demonstrated that chemical communication between neighbouring or more distant cells does not just involve endocrine and neurosecretory cells, but also unexpectedly tissues and organs such as the heart or the adipose tissue (e. g. the leptin signalling pathway). Comparative endocrinology could show that molecular components of hormonal systems represent signalling networks that are generally used during cellular communication processes and the differentiation of cell types during ontogenesis. Some of their functions are evolutionarily conserved, others not, as dissussion on steroid hormones and the prolactin signalling pathway will demonstrate.
