A practical route to the C24-C34 segment of FK 506 via (R)-(+)-3-cyclohexene-1-carboxylic acid (5). Key steps in the synthesis include asymmetric Diels-Alder and aldol reactions and enantioselective hydrolysis of a diester catalysed by pig liver esterase.
