Ropinirol - ein neuer Dopaminagonist

Zusammenfassung

Ropinirol ist ein Dopaminagonist der zweiten Generation mit selektiver D₂/D₃-Rezeptoren-Wirkung. Da es sich um eine nichtergoline Struktur handelt, ist ein besseres Nebenwirkungsprofil zu erwarten. Ropinirol weist in den Stadien Hoehn und Yahr I und II eine gleich gute Wirkung wie Levodopa auf, es ist Bromocriptin dagegen in allen Stadien überlegen. Besonders hervorzuheben ist seine geringe Induktion motorischer Fluktuationen. Neben Bromocriptin ist es als einziger Dopamin-agonist in der Monotherapie zugelassen aber auch in der Kombinationstherapie hoch potent.

Ropinirol is a dopamine agonist of the second generation with highly specific D₂/D stimulation. Since it is a non-ergoline structure there are less side effects such as fibrosis or Raynaud's syndrome. Ropinirol was as effective as levodopa when patients were treated who fulfilled the criteria of stage I and II of Hoehn and Yahr. In all HY stages ropinirol was more effective than bromocriptine. Ropinirol indices almost no motor fluctuations of hyperkinesias. Besides bromocriptine ropinirol is the only dopamine agonist permitted for monotherapy in Germany. It is also highly potent in combination therapy.
