The Distribution of ¹³¹I Labeled Diethylstilbestrol Diphosphate in Animals and Men with Observations on its Potential Use as a Scanning Agent of the Prostate^[*]

Summary

The distribution of ¹³¹I-labeled diethylstilbestrol diphosphate in the tissues of mice was found to be qualitatively similar to that of ¹⁴C-labeled and ³²P-labeled diethylstilbestrol diphosphate. A large percentage of the material is accumulated in the liver, from which it is excreted both via the bile and the gastrointestinal tract and kidneys. The concentration of the drug varied little, and tended to be lowest, in the prostate gland, testes, and adjacent tissues. The thyroid showed no large accumulation of radioiodide from the compound.

The distribution of the drug in dog tissues was qualitatively similar to that in mice tissues. The concentration of the drug in a dog’s prostate was delineated by scanning after the gland was located by palpation. The same region of the prostate was later rescanned after care had been taken to expose the prostate and to cover the bladder with sufficient lead.

The concentration of DES-2P-I* in human prostatic and testicular tissue was found to be similar to that previously reported for diethylstilbestrol and ³²P-labeled diethylstilbestrol diphosphate.

The possibility exists that the radiation from the labeled drug deposited in the prostate might affect tumor cell growth despite the very low concentration.

Scanning of the prostate is complicated by the accumulation of radioactivity in the bladder and colon.

Les auteurs ont trouvé la distribution du diéthylstilbestrol-diphosphate marqué au ¹³¹I dans le tissu des souris qualitativement pareille à celle du diéthyl-stilbestroldiphosphate marqué au ¹⁴C ou ³²P. Un pourcentage élevé du matériel se fixait dans le foie d’où il fut excrété par la bile ainsi que par les voies gastro-intéstinales et les reins. La concentration de la substance ne variait que peu. Elle était la moindre dans la prostate, les tésticules et le tissu adjacent. La glande thyroïdienne n’a pas montré une fication importante du radioiode du composé.

La distribution de la substance dans le tissu des chiens était qualitativement pareille à celle chez les souris. Après que la prostate du chien était localisée par palpation, la concentration de la substance fut tracée au moyen de scintigraphic Plus tard, la même région prostatique fut retracée, cette fois-ci la vessie étant couverte par le plomb.

La concentration du DES-2 P marqué au ¹³¹I dans le tissu prostatique et tésticulaire humain s’est montrée pareille à celle du DES et du DES-2 P marqué au ³²P auparavant communiquée.

Il existe la possibilité que l’irradiation de la substance marquée fixée dans la prostate puisse affecter la croissance des cellules cancereuses malgré la concentration assez moindre.

L’examination scintigraphique de la prostate est compliquée ce qui s’explique par la présence de radioactivité dans la vessie et le colon.

Die Verteilung des ¹³¹J-markierten Diaethylstilboestroldiphosphats in den Geweben der Maus ist qualitativ ähnlich jener des ¹⁴C- und ³²P-markierten Di-aethylstilboestrol-diphosphats. Ein hoher Prozentsatz der Verbindung wird in der Leber gespeichert und von dort sowohl *uuml;ber die Galle und den Verdauungs-trakt als auch über die Nieren ausgeschieden. Die Konzentration der Verbindung zeigt geringe Schwankungen und schien am geringsten in der Prostata, den Testis und deren benachbarten Geweben zu sein. Die Schilddrüse zeigte keine hohe Speicherung von Radiojod aus der Verbindung.

Die Verteilung der Verbindung in den Geweben des Hundes war qualitativ ähnlich jener in den Geweben der Maus. Die Konzentration der Verbindung in der Prostata eines Hundes wurde szintigraphisch festgehalten, nachdem die Lage der Drüse durch Palpation festgestellt worden war. Dieselbe Region der Prostata wurde nochmals szintigraphiert, nachdem die Drüse chirurgisch dargestellt und die Harnblase mit genügend Blei abgeschirmt worden war.

Die Konzentration von Diaethylstilbestrol-diphosphat im Gewebe der menschlichen Prostata und Testis war ähnlich jener, die früher für Diaethyl-stilboestrol und ³²P-markiertes Diaethylstilbestrol-diphosphat beobachtet wurde.

Es besteht die Möglichkeit, daß die Strahlung der in der Prostata gespeicher-ten markierten Verbindung das Wachstum von Tumorzellen trotz ihrer sehr geringen Konzentration beeinflussen könnte.

Die Szintigraphie der Prostata wird kompliziert durch die Ansammlung von Radioaktivität in der Harnblase und im Kolon.
