GnRH-Agonisten in der Reproduktionsmedizin beim Kleintier – eine Übersicht

Zusammenfassung

Kurzwirksame GnRH-Agonisten können beim Hund bei längerfristiger, täglicher Gabe erfolgreich zur Läufigkeitsinduktion eingesetzt werden. GnRH-Slow-release-Implantate bedingen nach einer initialen Stimulation eine Downregulation der hypophysären GnRH-Rezeptoren. Daraus resultiert eine temporäre Unterbindung der Produktion von Sexual-steroidhormonen, wodurch die Gonadenfunktion reversibel ausge-schaltet wird. Wenngleich das verfügbare Präparat Suprelorin^® ausschließlich für die Erzielung einer temporären Unfruchtbarkeit bei Rüden zugelassen ist, gibt es weitere Anwendungsmöglichkeiten von GnRH-Implantaten, bei denen eine Umwidmung sinnvoll erscheint. So kann vor Durchführung einer Gonadektomie beim Rüden das Auftreten kastrationsbedingter Nebenwirkungen geprüft werden. Ferner liegen positive Erfahrungen bei der Therapie der benignen Prostatahyper-plasie, von Adenomen der hepatoiden Drüsen und der so genannten Alopezie X vor. Der Einsatz bei der präpubertären Hündin dient dem Ziel der Verzögerung des Pubertätseintrittes. Bei adulten Hündinnen steht als Indikation die Ausschaltung des Zyklus im Vordergrund. Der günstigste Zeitpunkt der Implantatapplikation scheint in der Phase hoher Gelbkörperaktivität im frühen Diöstrus zu liegen. Dennoch besteht das Risiko der Läufigkeitsinduktion. Als neuartiger Therapieansatz ist die Verwendung eines GnRH-Agonist-Implantats bei kastrationsbedingter Harninkontinenz zu nennen. Sowohl bei Katze und Kater als auch bei männlichen Frettchen eignet sich der Einsatz von GnRH-Slow-release-Implantaten zur hormonellen Kastration. Eine weitere Indikation stellt die Hyperplasie und die tumoröse Entartung der Nebennierenrinde beim kastrierten Frettchen dar.

Summary

Short acting GnRH agonists can be successfully used in the dog for estrus induction after daily application. Following an initial stimulation, slow release GnRH agonist implants cause a downregulation of pituitary GnRH receptors. Due to the blockade of sex steroids a temporary reversible hormonal castration is achieved. Although the licensed indication for the GnRH implant Suprelorin^® is induction of temporary infertility in male dogs, there are several other indications like testing for possible castration-related side effects before performing gonadectomy, benign hyperplasia of the prostate, of adenomas of the hepatoid glands, and so called alopecia X. The use in the prepubertal bitch is for the delay of puberty. The main indication in adult bitches is prevention of estrus. For this the optimum time for implant application seems to be early diestrus with high luteal activity. Nevertheless the risk of estrus induction has to be taken into consideration. GnRH implants can also be used for treatment of urinary incontinence in neutered bitches. Moreover, hormonal castration is successfully achieved in tomcats and queens as well as in ferrets. Further indications for application of a slow release GnRH implant are hyperadrenocorticism or adrenal neoplasia in castrated ferrets.
