Incorporation of ¹³¹Iododeoxyuridine into the DNA of Tumourbearing Rats after Partial Synchronization as a Tool in Scintigraphic Tumour Localization

Après la synchronisation partielle avec l’Hydroxurée (HU), l’incorporation in vivo de ¹³¹Iododesoxyuridine (¹³¹IUdR) dans les tumeurs ainsi que dans dix organes des rats a été examinée. La synthèse de l’ADN ne comptait que 5% de la valeur normale, ceci pendant huit heures et demie. (La dose de HU appliquée six fois, à raison de 50 mg/kg du poids du corps). 90 minutes après la dernière application, des animaux par groupes de six furent traités avec du ¹³¹IUdR dans un délai de 30 minutes. 48 heures après le traitement avec du ¹³¹IUdR, les animaux ont été tués et l’activité du ¹³¹IUdR dans les tumeurs et les organes différents a été mesurées. Le degré de synchronisation ainsi que le maximum de fixation montraient des différences dans des différents organes. (Cerveaux et sang pas d’augmentation; iléum, foie, rate: facteur 1,5—1,8; poumon, tumeur, muscle, os: facteur 2,1—2,5; estomac, reins: facteur 3,6—4,5).

Les images scintigraphiques par le ¹³¹IUdR des rats nonsynchronisés et des rats partiellement synchronisés ont revelées que les tumeurs dans la région pulmonaire des rats peuvent être mieux demonstrées par une synchronisation partielle.

Summary

The rates of incoporation in vivo of ¹³¹Iododeoxyuridine (¹³¹IUdR) into the tumour and into 10 organs of tumour-bearing rats was investigated after partial synchronization with hydroxyurea (HU). The rate of DNA synthesis was reduced to 5% of the normal value over aperiod of 8.5 h by six successive injections of HU (50 mg/kg body weight). Commencing 1.5 h after the last HU injection, groups of 6 animals were injected with ¹³¹IUdR at intervals of 0.5 h. The animals were killed 48 h after this treatment and ¹³¹IUdR activity in the individual organs and in the tumour was determined. The results showed that the maximal uptake of ¹³¹IUdR after partial synchronization varied from organ to organ (not increased: brain, blood; increased by a factor of 1.5-1.8 duodenum, liver, spleen; increased by a factor of 2.1-2.5 lung, tumour, muscle, bone; increased by a factor of 3.6-4.5 stomach, kidney). Moreover, the time to reach maximal incorporation after HU treatment is different according to the tissue under investigation.

The use of ¹³¹IUdR as a tumour-localizing agent in scintigraphy of tumourbearing rats previously injected with HU showed that this method of partial synchronization clearly facilitates the detection of tumours in the lung area.

Es wurde der in vivo Einbau von ¹³¹Joddesoxyuridin (¹³¹JUdR) in den Tumor und in 10 Organe von Ratten nach vorheriger Teilsynchronisation mit Hydroxyharnstoff (HU) untersucht. Die DNS-Syntheserate wurde für 8,5 h auf 5% des Nöi-malwertes gedrückt (6malige HU-Gabe von 50 mg/kg Körpergewicht). Beginnend 1,5 h nach der letzten HU-Gabe wurde jeweils 6 Tieren im Abstand von 0,5 h ¹³¹JUdR appliziert. 48 h nach der Applikation wurden die Tiere getötet und de JUdR-Aktivität in den einzelnen Organen und im Tumor bestimmt. Dabei wurde in den einzelnen Geweben ein unterschiedlicher Synchronisationsgrad (Gehirn, Blut: keine Einbausteigerung; Dünndarm, Leber, Milz: Faktor 1,5—1,8; Lunge, Tumor, Muskel, Knochen: Faktor 2,1—2,5; Magen, Niere: Faktor 3,6— 4,5) sowie eine unterschiedliche zeitliche Lage der Einbaumaxima beobachtet.

Szintigraphische Aufnahmen mit ¹³¹JUdR von unsynchronisierten und teilsynchronisierten tumortragenden Ratten zeigten, daß durch Teilsynchronisation eine deutlich höhere Nachweisempfindlichkeit von Tumoren im Lungenbereich von Ratten erreicht werden kann.
