Significance
The first solid-phase synthesis of praziquantel via the Pictet-Spengler reaction was demonstrated. The polymer-supported linear acetal 1 was prepared from polyethylene glycol (PEG)-grafted hydroxymethyl polystyrene. The linear acetal 1 left the support as an acyliminium ion, which underwent a Pictet-Spengler reaction to generate praziquantel.
