Significance
Phorboxazole A is one of the most cytotoxic agents known. This synthesis, the fifth reported to date, was accomplished in 37 steps for the longest linear sequence, in 0.4% overall yield. For the Pd-catalyzed intramolecular alkoxycarbonylation that was used to construct fragments B and D, see: M. F. Semmelhack, N. Zhang J. Org. Chem. 1989, 54, 4483-4485.
