Solid-Phase Total Synthesis and Structure Proof of Callipeltin B

Significance
A solid-phase total synthesis and structure revision of cytotoxic, cyclic heptadepsipeptide, natural product callipeltin B, whose originally proposed structure was incorrect, were achieved. Three isomers 1a-c were synthesized (15%, 14% and 15% overall yields, respectively) in 11 steps from Tentagel-based TG Sieber amide resin 2. From comparison of the 1H NMR spectra of 1a-c with those of callipeltin B, the authors concluded that the correct structure of callipeltin B is 1c.
