Significance

(-)-Magnofargesin is an antagonist of platelet-activating factor (PAF), a phospholipid involved in numerous biological events. Inhibitors of PAF are of interest for the treatment of asthma. This synthesis is notheworthy for (1) the 1,5-C-H insertion of an alkylidinecarbene to generate a dihydrofuran, and (2) the use of a vinylsulfone as a fulcrum with which to install the appropriate functionality.
